Quick question

Realistically speaking, would mounting a sub using 6/8 holes affect the sound substantially over using all the holes?

As long as they're tightly secured and no air escapes from around the sub itself i don't imagine so.

use all screw hole openings, by not doing this the sub is not fastened to the baffle properly, when you look at the woofer mounting holes notice they are the exact same distance from one another there is a reason for this, use all 8 "genericmoniker"

i personally use all holes, but have been in a situation b4 when i couldnt use all, i used 6 i think, i did notice a bit of air leakage using 6 for my application, but took care of it with foil tape
