depends on the power, but realistically (and I'll get slaughtered for this by the SPL knuckledraggers) 1/2" should be fine for any daily driving application, as long as it's properly braced, and the power is kept under 1000 watts

One problem with anything smaller than 3/4" MDF is that it is harder to screw with out splitting it like a crazy. I think 3/4" is key for any sub/ amp combo, regardless of power. If you want 1/2" maybe look at baltic birch plywood, will not split but costs a pretty penny.
And yes the 1/2" will flex alot, and give you a bad sounding enclosure, and sloppy bass. Like everyone said you would need to brace it to make it solid.
